Rubber plug bioburden test method 1.Principle

The purpose of this test is to detect all aerobic plants and fermented materials and soil residues adhering to the gel plug by means of membrane filtration and to culture these filters.

1.Principle The purpose of this test is to detect all aerobic plants and fermented materials and soil residues attached to the rubber plug by means of membrane filtration,and to culture these filters.

2.Test Procedure 2.1 Preparation of Leachate The leachate was a 0.9%NaCl solution(with distilled water)and 1 ml of a nonionic active agent was added.

The above leachate was poured into a suitable glass container to a mark of 400 ml,and then the container was placed in an autoclave and kept at 121°C for 30 minutes for thorough sterilization.

Note:This experiment requires two glass containers,one for filtering aerobic plant residues adapted to neutral temperatures and the other for filtering the ferment and soil residues.
